About Routine/Basic Semen Examination
Routine/Basic Semen Examinations: Laboratory examination to assess quality and characteristics of semen. It is an assessment of the quality of semen and includes both the physical properties of semen as well as microscopic properties of the semen (sperm). Molecular Diagnostics and Therapy leverages cutting-edge laboratory technology to deliver accurate and reliable semen analysis results to aid with your assessment and treatment plan for fertility issues.
What is a Routine/Basic Semen Examination?
A Routine/ Basic Semen Examination includes an analysis of semen volume, liquefaction time, viscosity, pH, sperm concentration (count), motility, morphology, vitality, white blood cells, red blood cells, infection, and other abnormalities. This test is frequently ordered in the evaluation of male infertility, males after vasectomy, or in the investigation of various reproductive issues.
Common Reasons for a Routine/Basic Semen Examination
Male infertility evaluation
Difficulty achieving pregnancy
Assessment of sperm count and motility
Evaluation after vasectomy
Suspected reproductive tract infection
Investigation of abnormal semen quality
Follow-up after infertility treatment
Pre-marital or reproductive health assessment
Conditions Evaluated
Male Infertility
Helps identify abnormalities affecting sperm production and reproductive function.
Low Sperm Count (Oligospermia)
Evaluates reduced sperm concentration that may affect fertility.
Poor Sperm Motility
Assesses the ability of sperm to move efficiently for successful fertilization.
Abnormal Sperm Morphology
Evaluates the size and shape of sperm, which can influence fertility.
Azoospermia
Determines whether sperm are absent in the semen sample.
Seminal Tract Infection
Detects the presence of white blood cells and other findings suggestive of infection or inflammation.

Test information: 3 Days abstinence required
Reporting: Within 6 hours*
Sexual abstinence for 2–7 days is recommended before a Routine/Basic Semen Examination to ensure accurate results.
The semen sample should be collected by masturbation directly into a clean, sterile, wide-mouthed container provided by the laboratory. Avoid using condoms unless a special semen collection condom has been provided.
Wash your hands and genital area with clean water and dry thoroughly before collecting the sample to reduce the risk of contamination.
Ensure that the entire semen sample is collected, especially the first portion, as it contains the highest concentration of sperm.
If the sample is collected at home, keep it at body temperature (20–37°C) and deliver it to the laboratory within 30–60 minutes of collection.
Inform the laboratory if any portion of the sample was lost during collection, as this may affect the test results.
Avoid alcohol, smoking, recreational drugs, and excessive heat exposure (such as hot tubs or saunas) for at least 24–48 hours before the test, if possible.
Inform your doctor about any medications, recent fever, illness, or previous fertility treatments that may influence the results.
Bring your doctor's prescription and any previous semen analysis or fertility-related reports, if available.
Arrive at the diagnostic centre 10–15 minutes before your scheduled appointment.
Routine/Basic Semen Examination is a simple, safe, and non-invasive laboratory test used to evaluate male fertility and overall reproductive health.
The patient is asked to provide a semen sample by masturbation into a sterile, wide-mouthed container in a private collection room at the laboratory. If the sample is collected at home, it should be transported to the laboratory within the recommended time while maintaining body temperature.
Once received, the sample is allowed to liquefy naturally before analysis, as recommended by standard laboratory protocols.
The laboratory specialists then perform a comprehensive examination of the semen, including volume, colour, appearance, liquefaction time, viscosity, pH, sperm concentration (count), sperm motility, sperm morphology, vitality, and the presence of white blood cells, red blood cells, or microorganisms.
Additional microscopic evaluation helps identify abnormal sperm cells, signs of infection or inflammation, and other factors that may affect male fertility.
The findings assist doctors in diagnosing male infertility, low sperm count (oligospermia), poor sperm motility, abnormal sperm morphology, azoospermia, reproductive tract infections, and other reproductive disorders.
The procedure is completely painless, requires no injections or anaesthesia, and the laboratory analysis is usually completed within a few hours.
The results are reviewed by experienced laboratory experts, and the report is generally available within 24 hours.
